Inhibition of aquaporins as a potential adjunct to breast cancer cryotherapy

Cryoablation is an emerging type of treatment for cancer. The sensitization tumors using cryosensitizing agents prior to enhances ablation efficiency and may improve clinical outcomes. Water efflux, which regulated by aquaporin channels, contributes cancer cell damage achieved through cryoablation. An increase in (AQP) 3 cryoprotective, whereas its inhibition augments cryodamage. present study aimed investigate (AQP1, AQP3 AQP5) gene expression cellular localization response cryoinjury. Cultured breast cells (MDA-MB-231 MCF-7) were exposed freezing induce RNA protein extracts then analyzed reverse transcription-quantitative PCR western blotting, respectively. Localization aquaporins was studied immunocytochemistry. Additionally, transfected with small interfering silence viability assessed the Sulforhodamine B assay. Cryoinjury did not influence AQPs, except a 4-fold AQP1 MDA-MD-231 cells.
